Can't believe nobody has started a best line thread yet.
The winner for me was simply -

John Locke "Sorry..."

For me, the best line was:

"We cant just shoot him like a dog! Would you shoot a dog?"
"No! I like dogs!"

"You caught me."

Everything out of Locke's mouth... starting with "No, I like dogs!".. John was great in this episode!

John Locke "Sorry..."

Yeah, that reminded me of "I was wrong" in "Live Together, Die Alone." Hee.

I loved when Claire followed Desmond and said "What the hell is going on Desmond", or something really close to that.

FINALLY someone was going get to some answers. Now everyone needs to start asking everyone else what the hell is going on.

What did Locke call the fence? "Sonic death weapon" or something? I LOLed at that.

It's weird having such a Charlie-heavy episode not completely zing-laden.

This doesn't really count as a line but I loved Charlies "G'day" note to Claire.

I hope the week that Sawer can't use nicknames end really quick as the comic relief has been removed.

I liked Kate's attitude when she said "and why am I not capable of understanding?" and then "Try me."

They weren't funny or anything. I just liked how she was getting snarky with Patchy, and rightfully so.

of course, everything Locke said, including:

Saied: I thought you didnt know anything about the c4
Locke: I stand corrected

I also loved Kate's little, "What the hell John?" eye shrug.

Locke and Sayid were funny this week. This kind of humor I enjoy way more than the machine gun Sawyer quips. Sometimes you can go to the well too often, and I think they have done that with Sawyer on two episodes this season. Still, some people never tire of Sawyer's nicknames....

Sayid to John:

Certainly not as infallible as the carvings on your magical stick

Insert massive amount of irony into above

"Thank you."

I agree. This line was awesome because he just froze there for a split second then gets hit with the Sonic Boom.

You could see the relief on his face when he said this to Locke.

Dannielle~Why don't we just use your C4 to blast the fence (or something to that effect)

John~Do you want to walk up there and stick it on?

John's comment about liking dogs was funny too.

Not a lot of funny lines in this episode but some that really touched me.

Christian: "There is hope and there is guilt, and believe me I know the difference."

Claire: "I'm not giving up on you either, Charlie. It's gonna be okay. You're gonna be okay. We'll get through this together."

The "No! I like dogs" was classic.

Ditto. And of course, everything Locke said cracked me up, but nothing moreso than: "Why else would I be here?"

Sayid continuing to snark about the Jesus Stick...Sayid For President!

And a personal favorite:

Christian: "There is hope and there is guilt, and believe me I know the difference."

That should be the Shephards' motto.

"sorry" and "how was i supposed to know the place was rigged with C4?" (something like that) ....fastforward to being caught with aforementioned C4.

"Sorry", but more importantly that priceless psycho look Locke gave while saying it

I liked Claire's "I just figured out a way off this island" and she runs off at lightning speed down the beach. I thought that was cute.
